Tissue culture laboratory




The plant tissue culture laboratory building covers the area of 1500sq.ft. consisting of plant cell culture media preparation room, hot room,store room, office room, plant culture growth room, inoculation room, Mist chamber for primary hardening, shade house for secondary hardening.Tissue culture laboratory is equipped with electronic balance. Deioniser, autoclaves, Laminar flow chamber, Incubator and Photoperiod timer and Air conditioner.
Tissue culture laboratory has 2 growing Rooms sufficient to culture 10000 plant cultures at any given time depending on the plant species under production.
The laboratory has 4 Lamina flow bench for 10 Technicians to work at any given time. The Lamina flow benches are specially designed in order to decrease the rejection due to contamination which ensures to deliver plants on time.
The facility has Steam sterlisers used for sterlising the Media and all other materials used in the clean area.
Banana is a globally important fruit crop with 97.5 million tones of production. In India it supports livelihood of million of people. With total annual production of 16.91 million tones from 490.70 thousand ha., with national average of 33.5 T/ha. Maharashtra ranks first in production with 60 T/ha. Banana contributes 37% to total fruit production in India.

Objectives
- Conducting innovative research work in the field of plant biotechnology, development of varieties of cereal and horticultural crops using biotechnological interventions and molecular characterization and improvement of orphan crops of Tamil nadu.
- Identification, characterization and utilization of local bio-diversity of banana aromatic and medicinal plant species present in Tamil Nadu for commercial purpose.
- Development of technopreneurship in the state to contribute to its economic and hence social status.
- Exploitation of advanced biotechnological tools and techniques to enhance income in rural livelihood sector in Tamil Nadu.
Agro climatic conditions
Banana is basically a tropical crop, grows well in temperature range of 13ºC – 38ºC with RH regime of 75-85%. In India this crop is being cultivated in climate ranging from humid tropical to dry mild subtropics through selection of appropriate varieties like Grandnaine. Chilling injury occurs at temperatures below 12ºC. The normal growth of the banana begins at 18ºC, reaches optimum at 27ºC, then declines and comes to a halt at 38ºC. Higher temperature causes sun scorching. High velocity wind which exceeds 80 km phrs damages the crop.
Brief about Plat Tissue Cultures

Plant tissue culture is a novel and innovative technique to grow high quality, disease free plants quickly and in a large quantity by culturing and maintaining plant cells or organs like leaves, stem, root, branch shoot tip, petals, anther and pollen in sterile, nutritionally and environmentally supportive conditions in vitro. This technique is especially useful when the planting material is scarce or on the edge of extinction as thousands of plants can be produced from a single selected plant. Plants prepared using tissue culture technique are true to type resembling the mother plant in all traits, disease free, contains uniform growth and can be obtained throughout the year. Moreover, harvesting period for crops prepared with tissue culture plantations is earliest than normal. So, cultivation using tissue culture plants improves farmers income. The crop growth is also uniform with almost zero crop loss due to the diseases, which consequently leads to more yield, and thereby more profit.
The technology has enormous potential for meeting the demands of both domestic and export market in terms of high quality planting materials.

Plant Tissue Culture Lab
We have a modern, well equipped state of the art production facility with an annual capacity of 2 million plants per annum. The laboratory is having 2030.43. Sq. Mtr build up area for the operations and it comprises of two main sections sterile area and non-sterile area. Sterile areas (Bio clean) are having a sterility level of class 10,000.
Non sterile area is having media preparation, plant washing, grading and packing area.
We also have state of the art plant hardening facility in 2985.29 Sq. Mtr. area
Media Preparation Facility
Our media room is well equipped with basic equipments like pH meter, weighing balance (electronic) , conductivity meter , distillation unit, etc.
Autoclaving
Our high capacity sterilizers are with Microprocessor based automatic process sequence control system. In addition to the standard use of steam at higher temperature as the sterilizing agent, this unit also employs the vacuum system for added advantage in the sterilization process.
After autoclaving the media, it is stored for 2-3 days to check microbial contamination before use.
Plant Inoculation Room
The activity of segregating and transferring cultured plant material is being done in our Plant Inoculation room. Fully controlled and sophisticated plant inoculation rooms with total 3 laminar flow benches with a seating capacity of 9 operators.
Growth Room
The growth room is designed to grow cultures with controlled light. Humidity and temperature. Culture racks are fitted with the fluorescent tube lights to ensure optimum availability of light for proper growth of the cultures. The rooms are fumigated periodically to keep the contamination under control. Each growth room has capacity to maintain different temperatures. Growth room area sterility is also maintained to control contamination.
Greenhouse
We have climate controlled greenhouses in an area of 500 sq mt for the primary hardening of 10000 plants/cycle. Green houses are maintained at desired temperature and humidity with the help of latest control panel
Plants are monitored regularly for their growth and presence of any disease or pest. Periodical and timely spraying of suitable chemicals are carried out with proper recording and reference.
House
We have 500 sq mt of the shade house area equipped with foggers and misters for the secondary, assures Utmost care is taken for selection of proper tray, potting mixture, during washing of plants, water for irrigation. All records pertaining to transfer of plants is maintained for tracking of movement of plants.
